ABSTRACT

OBJECTIVE:To ex plore the effects of solamargine on the growth and apoptosis of human hepatocarcinoma cells HepG2 and its underlying mechanism. METHODS :The effects of 0(blank group )-12 μmol/L solamargine treatment of 24,48 h on survival rate of HepG 2 cells were investigated. The effects of 0(blank group ),6 μmol/L solamargine treatment of 10 days on cell clone formation were also investigated. The effects of 0(blank group ),4,6,8 μmol/L solamargine for 24 h on the apoptotic rate of cells,mRNA expression of Bcl- 2,Bax and caspase- 3, protein expression of Bcl- 2 and cleaved caspase- 3 as well as ratio of p-AMPKα to AMPKα were all tested. The effects of AMPK inhibitor as compound C on the protein expression of AMPKα and Bcl- 2 in cells were investigated after treated with 6 μmol/L solamargine for 24 h. RESULTS :Compared with 020-39318678。E-mail:wujingjing6028@gzucm.edu.cn blank group ,1-12 μ mol/L solamargine for 24,48 h could significantly decrease the survival rates of cells (P<0.05)in a concentration-dependent manner ;IC50 of them were 8.310 and 7.996 μmol/L,respectively;the rate of cell clone formation was decreased significantly after treated with 6 μmol/L solamargine for 10 days(P<0.05). The apoptotic rate of HepG 2 cells,mRNA expression of Bax and caspase- 3,protein expression of cleaved caspase-3(except for 8 μmol/L)as well as ratio of p-AMPKα to AMPKα(except for 8 μmol/L)were all increased significantly after treated with 6,8 μmol/L solamargine(P<0.05);mRNA and protein expression of Bcl- 2 were decreased significantly (P< 0.05);the changes of some indexes were in a concentration-dependent manner. The compound C could inhibit protein expression of AMPKα,and reverse the inhibitory effect of solamargine on Bcl- 2 protein. CONCLUSIONS :Solamargine can inhibit the proliferation of HepG 2 cells and induce apoptosis ,the mechanism of which may be associated with activating AMPK signaling pathway.

ABSTRACT

Objective Solamargine (SM), with its anti-inflammatory and anti-tumor effects, inhibits the proliferation and promotes the apoptosis of various tumor cells. This study was to investigate the effects of SM on the proliferation and apoptosis of human esophageal cancer KYSE150 cells and its action mechanism. Methods We treated KYSE150 cells with SM at the concentrations of 0 (the blank control group), 2, 4, 6 and 8 μmol/L for 24 hours. Then, we observed the morphological changes of the cells under the inverted microscope, detected their proliferation and apoptosis by MTT assay and flow cytometry respectively, and determined the expressions of the classical NF-κB signaling pathway-related proteins NF-κB, p-NF-κB, IKKα, IKKβ, IkBα and p-IkBα) and apoptosis-related proteins Bax, caspase-3, cleaved caspase-3 and Bcl-2 in different groups of the cells by Western blot. Results Compared with the blank control, the inhibition rate of the proliferation of the KYSE150 cells in the 2, 4, 6 and 8 μmol/L SM groups was increased significantly in a concentration-dependent manner (0 vs [15.03 ± 0.15]%, [47.94 ± 1.74]%, [68.72 ± 0.47]% and [77.51 ± 1.70]%, P<0.05), and so was the apoptosis rate ([8.17 ± 0.51]% vs [14.50 ± 0.73]%, [18.57 ± 2.08]%, [65.10 ± 10.88]% and [81.55 ± 5.48]%, P<0.05). The expression of the apoptosis-related protein Bax in the SM treated cells was up-regulated, those of Bcl-2, IKKα, IKKβ and p-IkBα down-regulated, and the activity of caspase-3 and cleaved caspase-3 promoted, all in a concentration-dependent manner, with statistically significant differences between the blank control and the 4, 6 and 8 μmol/L SM groups (P<0.05). Statistically significant differences were also found in the expressions of NF-κB, p-NF-κB and IkBα between the blank control and the 6 and 8 μmol/L SM groups (P<0.05). Conclusion Solamargine can significantly inhibit the proliferation and promote the apoptosis of KYSE150 cells, probably by suppressing the classical NF-κB signaling pathway.

ABSTRACT

ABSTRACT Leishmania infantum is an etiologic agent of visceral leishmaniasis. This disease is a neglected disease that can be fatal if not treated and additionally, the few therapeutic option present several drawbacks, including difficult route of administration and toxicity, which turn the search for new therapeutic alternatives necessary. Herein, we evaluated the leishmanicidal in vitro activity of the solanum extract from Solanum lycocarpum A. St.-Hil., Solanaceae, and the isolated alkaloids solasodine, solamargine and solasonine against promastigotes and intracellular amastigotes of L. infantum. Solasodine (IC50-pro = 4.7 µg/ml; IC50-ama = 10.8 µg/ml) and solamargine (IC50-pro = 8.1 µg/ml; IC50-ama = 3.0 µg/ml) exhibited interesting leishmanicidal ativity. Solasonine was approximately four-times (Selective Index 3.7) more selective to the parasite than to the host cells. This data suggest that solasonine might be considered as a potential drug candidate for leishmaniasis treatment.

ABSTRACT

The majority of penile cancers develop from squamous cells within the skin resulting in a non-melanoma form of skin cancer, squamous cell carcinoma. Squamous cell carcinomas can develop anywhere on the penis, most appear on the foreskin in men who have not been circumcised or on the glans. Various types of squamous cell carcinomas ranging from Bowen’s disease, erythroplasia of Queyrat and Buschke- Lowenstein tumour have been identified on the penis. When detected early, these types of cancer can be successfully treated. However, the current treatment procedures have the potential for disfigurement and dysfunction of male genitalia, with associated psychological distress. A relatively new class of antineoplastic agents, consisting of the solasodine rhamnosides, solamargine and solasonine, are very effective and safe for the treatment of a variety of skin cancers and show promise for the treatment of internal cancers. Here, three cases of distinct squamous cell carcinomas on the penis, Bowen’s disease, erythroplasia of Queyrat and Buschke-Lowenstein tumour are reviewed which were successfully treated with a standard mixture of solasodine rhamnosides.
